Mathematical modeling of eIF5B-mediated non-canonical translation initiation as a chemotherapeutic target Under hypoxic conditions, which is observed in cancer cells, cap-dependent translation initiation is disrupted via the phosphorylation of eukaryotic initiation factor 2- (eIF2). However, cancer cells can exploit a non-canonical pathway that replaces eIF2 with eIF5B for initiator tRNA delivery to the ribosome, resulting in the synthesis of antiapoptotic proteins such as XIAP. Mathematical models were derived and used to identify several potential therapeutic targets in this non-canonical mechanism. |

Expanding the repertoire of self-amplifying RNA vectors: an emerging frontier in mRNA vaccines for infectious diseases Self-amplifying RNA (saRNA) results in prolonged and higher expression of encoded gene compared to standard mRNA. We designed saRNA vectors from alphaviruses. We showed that by tuning the vector design, we modulate the magnitude and duration of protein expression ex vivo and in vivo and influence the humoral and cellular immunity generated by saRNA vaccines using COVID-19 as a model system. |

Tuning the ykkC-I riboswitch: insights into potential effects of metal ion-induced allostery through MD simulations Riboswitches exhibit high ligand specificity and potential as drug targets. This study focuses on ykkC (guanidine-I) riboswitches, the sole receptors for guanidinium cations, using long-timescale molecular dynamics simulations. Key findings reveal how guanidinium ions and metal cations drive structural transitions, modulate ligand-binding dynamics, and influence RNA regulation. These offer insights into RNA-based regulation and potential applications in drug discovery and biosensing. |

The Translation Machinery in Health and Disease GRC will highlight new molecular insights into translation including control during normal processes and how their dysregulation contributes to a wide array of diseases. The GRC on RNA Editing is the only international meeting dedicated exclusively to RNA editing and RNA modifications. The 2025 meeting is subtitled “Biological Mechanisms and Applications of RNA Editing and Modification” and will showcase the breadth of the newest advancements in this rapidly growing field. RNA editing and modification are highly abundant phenomena, occurring in almost all RNAs, in all cells, and in all organisms. These relatively simple molecular events carry far-reaching biological consequences, and our knowledge about them is expanding quickly. Importantly, many biological roles of RNA editing and modification are actively being uncovered, and these range from the control of gene expression, adaptation to stress, viral, microbial and parasitic infectivity, organismal development, brain function, innate and adaptive immunity, inherited diseases, cancer and more. Based on a strong foundation in mechanistic and functional investigations on RNA editing and modification enzymes, our research field has now entered a new and exciting era where a variety of applications are emerging that utilize RNA editing and modifications as tools for research and health. The 2025 meeting will showcase these novel applications in the areas of therapeutics that alter the transcriptome and proteome, more effective mRNA vaccines, among others. A goal of this meeting is to facilitate the translation of academic discoveries in the area of RNA editing and modification into platforms that advance biotechnology and health. Overall, the 2025 GRC on RNA Editing will accelerate discovery by inspiring research into biological mechanisms, by fostering knowledge translation, by building a strong, diverse, and inclusive community and, most importantly, by developing the next generation of creative scientists.

The goal of the NCI RNA Biology Initiative is to establish a collaborative environment that promotes the swift exchange of information and expertise regarding the structure, function, and biological significance of RNA. Our objective is to leverage this collective knowledge to advance the development of novel diagnostics and therapies. We actively foster synergistic collaborations among NCI and NIH intramural investigators, as well as between our researchers and the broader RNA community.
The symposium offers the opportunity to learn more about the current status of RNA biology in development and disease, share research, network with leading figures in the field and discuss the use and implications of these advances for clinical applications.
